چکیده
INTRODUCTION The bioluminescent bioreporter integrated circuit represents a new advance in the development of whole-cell biosensors. These devices consist of a genetically engineered bioreporter organism interfaced with an integrated circuit (Fig. 1). The bioreporter is engineered to luminesce when a targeted substance is encountered, while the circuit is designed to detect the luminescence, process the signal, and communicate the results. The chief advantage of this approach is that the entire sensor, including all signal-processing and communication functions, can be produced as a single-chip, low-power, rugged, inexpensive device. We envision these devices being used in a variety of distributed sensing and environmental control systems. Future work will focus on using more information processing capabilities of the cells to create highly functional sensing/computing/actuating devices.

A bioluminescent bioreporter integrated circuit (BBIC) is a novel whole-cell biosensor that combines the environmental monitoring capabilities of genetically engineered bioluminescent micro-organisms (bioreporters) with optical application-specific integrated circuits. A BBIC device consists of bioreporters sustained within a micro-environment, the integrated circuit microluminometer, and a lig...

The luxA, B, C, D, and E genes from Photorhabdus luminescens were cloned and functionally expressed in Saccharomyces cerevisiae to construct a bacterial lux-based yeast bioreporter capable of autonomous bioluminescence emission. The bioreporter was engineered using a series of pBEVY yeast expression vectors that allowed for bi-directional constitutive or inducible expression of the individual l...

Living cells of the lux-based bioluminescent bioreporter Pseudomonas putida TVA8 were encapsulated in a silica hydrogel attached to the distal wider end of a tapered quartz fiber. Bioluminescence of immobilized cells was induced with toluene at high (26.5 mg/L) and low (5.3 mg/L) concentrations. Initial bioluminescence maxima were achieved after >12 h. One week after immobilization, a biofilm-l...
